[Effects of miR-144-3p on Proliferation, Cell Cycle and Apoptosis of K562 Cells]

Abstract

Objective: To investigate the effects of miR-144-3p on cell proliferation, cell cycle and apoptosis of blast phase chronic myelogenous leukemia (CML) K562 cells.

Methods: K562 cells were cultured in vitro and mimics negative control, hsa-miR-144-3p mimics, inhibitor negative control and miR-144-3p inhibitor were respectively transfected into K562 cells with transfection reagents. The cells were divided into five groups including blank control, mimics negative control, miR-144-3p mimics, inhibitor negative control and miR-144-3p inhibitor. After transfection, the cell proliferation activity was detected by CCK-8 assay. The cell cycle distribution and apoptosis were detected by flow cytometry.

Results: Compared with the blank control and mimics negative control groups, the proliferation rate of miR-144-3p mimics group was significantly decreased (P<0.05), the proportion of S phase cells was markedly increased (P<0.05), while the proportion of G1 phase cells was obviously decreased (P<0.05), and the apoptosis rate was significantly increased (P<0.05). Compared with the blank control and inhibitor negative control groups, the proliferation rate of miR-144-3p inhibitor group was obviously increased (P<0.05), the proportion of S phase cells was markedly decreased (P<0.05), while the proportion of G1 phase cells was obviously increased (P<0.05), and the apoptosis rate was significantly decreased (P<0.05).

Conclusion: miR-144-3p can inhibit the proliferation and promote apoptosis of K562 cells, affect the cell cycle, and block K562 cells in S phase, which indicates that miR-144-3p is involved in the cell cycle activity of CML during blastic phase.

目的: 研究miR-144-3p对慢性髓性白血病急变期K562细胞增殖、周期和凋亡的影响.

方法: 体外培养K562细胞,通过转染试剂分别将模拟物阴性对照、miR-144-3p模拟物、抑制物阴性对照、miR-144-3p抑制物转染入K562细胞。将细胞分为空白对照、模拟物阴性对照、miR-144-3p模拟物、抑制物阴性对照和 miR-144-3p抑制物共5组。采用 CCK-8 法检测细胞增殖,使用流式细胞术检测细胞周期和凋亡情况.

结果: 与空白对照、模拟物阴性对照组比较,miR-144-3p模拟物组细胞增殖率明显减低(P<0.05),S期细胞明显增加(P<0.05),G1期细胞明显减少(P<0.05),细胞凋亡率明显升高(P<0.05);与空白对照、抑制物阴性对照组比较,miR-144-3p抑制物组细胞增殖率明显升高(P<0.05),S期细胞明显减少(P<0.05),G1期细胞明显增加(P<0.05),细胞凋亡率明显下降(P<0.05).

结论: miR-144-3p能够抑制K562细胞增殖、促进凋亡,影响细胞周期,将K562细胞阻滞在S期,表明miR-144-3p参与了慢性髓性白血病急变期细胞周期活动.
