[Impact of sequential targeted therapy on progression-free survival in metastatic renal cell carcinoma]

Abstract

Objective: To evaluate the impact of sequential targeted therapy including vascular endothelial growth factor receptor tyrosine kinase inhibitors (VEGFR-TKI) and everolimus on progression-free survival (PFS) in metastatic renal carcinoma.

Methods: A total of 17 patients of metastatic renal cell carcinoma from Department of Kidney Cancer and Melanoma, Peking University Cancer Hospital since January 2008 until April 2014 were enrolled into this retrospective study. They took at least 2 lines of VEGFR-TKIs and everolimus. The data of total PFS and PFS of each therapy were extracted and analyzed.

Results: The sequential targeted therapies affected total PFS (18.0 vs 46.0 months, Kaplan-Meier survival analysis, P = 0.015) with a statistical dislike of VEGFR-TKI/VEGFR-TKI/mTOR inhibitor (TTM) over VEGFR-TKI/mTOR inhibitor/VEGFR-TKI (TMT) , HR = 5.44 (Cox regression analysis, 95% CI 1.18-25.06).

Conclusion: TMT may prolong the total PFS of metastatic renal cell carcinoma.
