Currently many biopharmaceuticals are in use due to their high efficacy and low adverse effects. For antibody drug formulation, liquid ones are being increasingly used. This review focuses on rational liquid antibody formulation development based on the proposed aggregation mechanism of antibody, considering colloidal and conformational stabilities. This review also describes the importance of assessment of physicochemical properties including second virial coefficient of antibody solutions, for the rational and efficient formulation development. This article is part of a Special Issue entitled: Recent advances in molecular engineering of antibody.
Keywords: Aggregation; Antibody; Colloidal stability; Conformational stability; Formulation; Virial coefficient.
