[An application of oligonucleotide-directed mutagenesis on making deletion mutation]

Abstract

By the method of oligonucleotide-directed mutagenesis, we made 27bp and 18bp deletion at junction region between alpha-factor signal sequence and alpha-hANP gene, alpha-factor signal sequence and alpha-IFN gene, respectively. Since the deleted region contains one HindIII site, the mutants without this site were selected. The result of DNA sequence analysis showed that the sequences of the mutants were the same as designed.
