Invariant natural killer T cells (iNKT cells) are a small subset of T lymphocytes that are equipped with various immunoregulatory cytokines and cytotoxic effector molecules. Immune responses can be modulated efficiently by their interaction with other cells of the innate and adaptive immune system. Also, iNKT cells can promote apoptosis of malignant cells. Therefore, iNKT cells play a particular role in infectious diseases, malignant diseases, autoimmunity and alloimmunity. After allogeneic hematopoietic cell transplantation, iNKT cells induce immune tolerance and promote graft-versus-tumor effects. Recent advances in automated cell processing under good manufacturing practice (GMP) conditions and genetic modifications of effector cells pave the way for clinical translation of iNKT cell therapy.
Invariante natürliche Killer-T-Zellen (iNKT-Zellen) sind eine kleine Subpopulation der T‑Lymphozyten, die mit einem erstaunlichen Repertoire an immunregulierenden Zytokinen und zytotoxischen Effektormolekülen ausgestattet sind. Diese befähigen sie einerseits zur Modulation von Immunantworten über die Interaktion mit anderen Zellen der angeborenen und adaptiven Immunität. Andererseits können iNKT-Zellen den Zelltod bösartiger Zellen induzieren. Somit spielen sie eine wichtige Rolle bei Infektionskrankheiten, Malignomen, Autoimmunität und Alloimmunität. Insbesondere im Kontext der allogenen hämatopoetischen Stammzelltransplantation entfalten iNKT-Zellen ihr Potenzial durch die Induktion immunologischer Toleranz und verbesserter Malignomkontrolle. Die jüngsten Fortschritte in der Zelltherapie hinsichtlich automatisierter Produktionsverfahren unter den Bedingungen der guten Herstellungspraxis („good manufacturing practice“, GMP) sowie die Möglichkeiten der genetischen Modifikation zur Steigerung der Effektivität ebnen den Weg für die erfolgreiche klinische Anwendung von iNKT-Zellen.
