Reverse transcriptase dideoxy sequencing of RNA

Methods Enzymol. 2013:530:347-59. doi: 10.1016/B978-0-12-420037-1.00020-8.

Abstract

During the course of studying the structure and function of RNA molecules and the proteins that they may encode, as well as studies of the expression of a given gene, there often arises a need to learn the nucleotide sequence of an RNA molecule. While the sequence of a particular RNA is ordinarily encompassed within a corresponding DNA sequence, RNA sequencing lends the advantage of giving the transcription initiation and termination sequences, and the sites where modifications from the original DNA sequence occur.
